GENETIC EFFECTS OF ENVIRONMENTAL CHEMICALS

Objective

1) STUDY OF MOLECULAR INTERACTIONS OF ENVIRONMENTAL MUTAGENS AND CARCINOGENS AT DNA LEVEL. 2) TO RELATE BIOLOGICAL CONSEQUENCES OF CHEMICAL MUTAGENS TO THE EXTENT AND SITE OF REACTION BETWEEN THE MUTAGENS AND DNA. 3) DEVELOPMENT AND VALIDATION OF METHODS TO IDENTIFY NON-GENOTOXIC CARCINOGENS. 4) DEVELOPMENT AND VALIDATION OF ASSAYS TO EVALUATE CHEMICALS CAPABLE OF INDUCING GENOMIC MUTATIONS.

1) AND 2) RELATIONSHIP BETWEEN IN VITRO AND IN-VIVO MUTAGENESIS: THE INDUCTION OF CHROMOSOME ABERRATIONS IN LYMPHOCYTES, HEPATOCYTES AND BONE MARROW CELLS, AFTER IN-VITRO OR IN-VIVO EXPOSURE TO MUTAGENS WITH A KNOWN DOSIMETRY WILL BE STUDIED, IN ORDER TO CORRELATE THE SAME GENETIC ENDPOINTS ON THE SAME CELL TYPE EITHER IN-VITRO OR IN-VIVO.

3) COMPARATIVE CHANGES IN THE TISSUES OF MICE TREATED IN-VIVO AND STUDIED IN-VITRO: CYTOGENIC ANALYSES WILL BE CARRIED OUT IN EPITHELIAL MOUSE CELLS IN-VIVO AND IN CULTURES OF THE SAME CELLS, AFTER TREATMENT WITH VARIOUS MUTAGENS AND TUMOUR PROMOTERS.

4) MICRONUCLEI IN-VITRO AND SOMATIC TISSUE IN-VIVO - ANEUPLOIDY IN MALE GERM CELLS;

I) EVALUATION OF CHROMOSOME NUMBER IN METAPHASES, THE PRESENCE OF MICRONUCLEI INTERPHASE HUMAN LYMPHOCYTES; ALSO IN SOMATIC CELLS OF DIFFERENT TISSUES OF RODENTS AFTER IN-VIVO TREATMENTS.

II) INDUCTION OF NUMERICAL ANOMALIES IN MOUSE GERM CELLS CAUSED BY THE SAME COMPOUNDS.
