RESEARCH ON TOXIC MATERIALS WITH THE MICROCALORIMETRY OF BACTERIAL METABOLISM

Objective



THIS RESEARCH PROJECT WILL :

- STANDARDIZE A SUBLETHAL MICROBIOLOGICAL ECOTOXICITY TEST, IN TAKING INTO ACCOUNT THE METABOLISM OF WELL DEFINED BACTERIAL STRAINS, FOR CHEMICAL SUBSTANCES WHICH WILL BE REGULATED BY COUNCIL DIRECTIVE 76/464/EEC;

- APPLY REFINED BIOPHYSICAL EXPERIMENTAL SYSTEMS IN THE DEVELOPMENT OF MICRO-BIOLOGICAL ECOTOXICITY TESTS, SUCH AS FLOW-MICROCALORIMETRY IN COMBINATION WITH POLAROGRAPHY, SPECTROFLUORIMETRY AND MEASUREMENTS OF THE OPTICAL DENSITY, NUTRIENT CONCENTRATION AND PH. THIS WILL IMPROVE THE SENSIBILITY OF QUANTITATIVE MEASUREMENTS OF METABOLIC RESPONSES OF MICROORGANISMS IN THE PRESENCE OF CHEMICAL POLLUTANTS. IN PARTICULAR "ENERGETIC CURVES" (TIME EVOLUTION OF DISSIPATED HEAT) UNDER CONDITIONS OF VARIOUS GRADES OF METABOLIC PERTURBANCE WILL BE ELABORATED AS SUBLETHAL INDICATOR FOR TOXIC EFFECTS.

- A MONTECARLO SIMULATOR WILL BE USED, IN ORDER TO EXTEND THE EXPERIMENTAL INFORMATION FOR VARIOUS BOUNDARY CONDITIONS AND FOR PREDICTING TOXICOLOGICAL EFFECTS.
