Research towards the total synthesis of antibiotic amipurymicin

Objective



Amipurimycin and Miharamycins are nucleoside antibiotics active against Pyricularia oryzae, the phytopathogenic fungi causing the rice blast disease. (display: AMIPURIMYCIN MIHARAMYCIN A: X=OH MIHARAMYCIN B: X=H) These compounds exhibit the following common structural features: * Same 2-amino purine aglycon * Closely related branched chain moieties * C-glycopyranosylated alpha-aminoacid substructures including 6-amino-6-deoxy-hepturonic acids. The third point appears of particular importance owing to the fact that the absolute configurations at C-6' remain to be established. The present project is aimed at the syntheses of novel nucleosides containing these 6-amino-6-deoxy hepturonicacid moiety as well as the 2-aminopurine base. These compounds will be evaluated against Pyricularia oryzae in order to determinate the structural features responsible for the biological activity. All the new compounds will be submitted to phytobiologic studies in the Centre de tests Rhtne Poulenc Agrochimie.
