Structural determination and development of a sitedirected mutagenesis system for the photosynthetic reaction centre from the purple bacterium rhodopseudomonas acidophila

Objective



The objectives of this proposal are to obtain a high resolutlon atomic structure for the photosynthetic reaction centre (RC) from the purple bacterium Rhodopseudomonas acidophila and subsequently to develop a site-directed mutagenesis system. The understanding of structure/function relationships in RCs are hindered by the lack of (1) a third structure for comparative analysis and (2) a stable mutagenesis system for Rhodospeudomonas viridis due to the inability of this species to grow chemoheterotrophically. The work program to meet the objectives aims to exploit the experience accumulated at the host institute concerning membrane protein crystallisation coupled with; the availability of a pre-existing genomic library, chemoheterotrophic growth of Rhodopseudomonas acidophila, and the ability to mobilise cloning vectors into this species. This proposal would also provide me with an unique opportunity to receive specialised training in the field of membrane protein crystallography.
