Activation and assembly mechanisms of the rhodococcus proteasome

Objective



Proteasomes are essential components of eukaryotic cells and have also been found in prokaryotes. They play a key role in intracellular protein degradation and are composed of related subunits which fall into two families, named a- and B-type subunits.
A general feature of all proteasome B-type subunits is a highly variable prosequence. We plan to explore the role of the B propeptide in the activation and assembly mechanisms of the Rhodococcus prokaryotic proteasome. It is an extremely useful research tool because it displays an intermediate complexity between the archaebacterial and eukaryotic complexes. A prime focus of our work will be to elucidate the mechanisms involved in the cleavage of the prosequence. The other major goal will be to investigate the potential role of the prosequence in targeting a B subunit to its a partner. These investigations will be performed by using multidisciplinary approaches combining recombinant DNA techniques, site-directed mutagenesis and electron microscopy.
