The synthesis of the galbulimima alkaloids himgravine and himbacine - potential therapeutic agents for alzheimer's disease

Objective



In the last decades isolation and screening of natural products has become a very important factor in finding new therapeutic agents. During this project a synthesis for the Galbulimima Alkaloids Himgravine 7 and Himbacine 8 should be developed. These Alkaloids should stimulate the release of acethylcholin and due to the cholinergic hypothesis should alleviate the symptoms for Alzheimer's disease. A retrosynthetic approach, as illustrated in scheme 1, shows that suitable building blocks can be prepared starting with substrates from the chiral pool like amino acids, lactic acid 1 as well as heterocycles 6 by using various standard organic reactions (C-C coupling reactions, protection and deprotection of functional groups and stereoselective synthesis). Modern analytical methods like HPLC, GC, NMR-, IR- and Masspectrometrie will be used to identify the intermediates and products.
