Mapping of systemic lupus erythematosus - sle - susceptibility genes by linkage disequilibrium in the unique Finnish population

Objective



The objectives of this proposal are to map susceptibility genes predisposing to SLE in the Finnish population, and to determine the general feasibility of linkage disequilibrium (LD) mapping strategies for the identification of complex disease susceptibility genes in a genetically homogenous population. A novel approach based on the special genetic features of the Finnish population, namely the extensive LD found between individuals, is proposed.
The project entails the systematic genotyping of DNA samples from over 200 Finnish SLE families with microsatellite markers to identify shared genomic segments of LD by analyzing human homologous regions containing candidate genes implicated in previous SLE studies and by performing a genome-wide search. Statistical analysis of the obtained genotype data for allele and haplotype associations with the disease in affecteds and controls will identify candidate SLE susceptibility gene regions.
