Molecular genetic analysis of colonization factors of lactobacillus

Research objectives and content
Colonization is thought to play an important role in determining probiotic properties of lactobacilli. The relationship between structure and function of two Lactobacillus adhesion proteins will be studied to increase our insight in mechanisms of colonization and to contribute in designing strains suited as vehicles mucosal immunization programmes. The first part of the project, determination of regions of S-layer proteins involved in recognition of receptors on the extracellular matrix, involves the construction and analysis of hybrid S-proteins comprising regions of L. crispatus (adhesion positive) and L. acidophilus ( adhesion negative) S-proteins.
Secondly, the structure-function relationship will be determined of an adhesion factor of L. fermentum showing strong similarity to virulence factors from pathogenic bacteria. A mutation analysis will be performed of regions of the protein known to be involved in adhesion of L. fermentum to pig gastric mucosa. The capacity to compete with pathogenic bacteria will be determined.
Finally, other Lactobacillus strains will be screened to determine the occurrence of adhesion proteins by using antibodies against these proteins and specific DNA probes. Their adhesion properties will be tested. Training content (objective, benefit and expected impact)
The main training content is concerned with the molecular biology of colonising lactobacilli. In this way, I will become familiarized with i) procedures of gene fusion and expression of hybrid proteins in homo- and heterologous hosts, ii) mutation analysis and adherence test in Lactobacillus, being all of them complementary to those I have already carried out. So, I could continue my professional development in lactic acid bacteria, previously focused on Lactococcus.
