The role of the tumour metastasis gene tiam1 in cell signalling and development

Objective



Research objectives and content The tumor metastasis gene Tiaml was identified through retroviral mutagenesis of T-lymphoma cells followed by screening for invasive phenotypes. This gene encodes a guanine nucleotide exchange factor for the Rho-family small GTPase Rac demonstrating a role for the Rho-family in tumor invasion and metastasis. The normal, non-oncogenic cellular context of Tiaml signalling through Rac has yet to be defined, although Tiaml expression in specific regions of the brain suggests roles in neuronal migration and maturation. Since preliminary data suggests that interacting proteins regulate Tiaml activity we will characterize partners for Tiaml through protein interaction assays. At the same time we will develop transgenic models to elucidate the role of Tiaml in brain tissue development through conditional expression of wildtype, constitutively active and dominant negative Tiaml mutants. This project will improve our understanding of Tiaml function in development and in cancer metastasis. It will demonstrate the utility of emerging techniques applied to investigate this important family of oncogenes. Training content (objective, benefit and expected impact) The training will familiarize me with contemporary techniques in cell biology and give me specialized knowledge in development of transgenic mice and their subsequent analysis. These are techniques which will complement my present expertise in molecular biology.
