Interaction of metal compounds of quinolone drugs with dna

Research objectives and content
The main subject of our research is the study of metal compounds which are the products of the interaction of metal complexes that exhibit anticancer activity with some of the quinolones drugs that are antibacterial agents. The purpose of this study is to synthesize new, less toxic and more effective chemotherapeutic agents. At the host Institute I am going to examine interaction of these compounds with synthetic and natural DNA by 1D and 2D (COSY, noesy) NMR techniques and Circular Dichroism (CD), as the action of many antitumor drugs is suggested to be performed through interaction with DNA. Our objective is to obtain results that will help as elucidate the metals' mode of action on DNA and how it can be affected from the quinolone drugs' presence and the base sequence.
Training content (objective,benefit and expected impact)
The host laboratory at the Department of Chemistry in the Bergen University, is one of the few laboratories in Europe that have all the required instruments for carrying out the experiments with synthetic oligonucleotides (e.g. Iarge NMR facilities, 600 MHz). Moreover the research group has a great experience in all the techniques required and Prof.E.Sletten is an authority on the field of interaction of metal ions and their complexes with DNA. This will not only help me carry out a basic part of my research Thesis that I could not complete otherwise but in addition I will have the opportunity to work with a well known research team in Europe.
