Identification of signalling pathway components for pathogenic development of ustilago maydis

Research objectives and content
U. maydis is a model organism for studies on plant pathogen fungi. The aim of this project is to characterise signalling pathways components that act in relation with Gpa3, a G alpha subunit of heterotrimeric G protein in U. maydis pathogenic development. Gpa3 controls the cAMP pathway which plays a fundamental function in pathogenicity. Gpa3 also regulates a second pathway involved in tumor formation. Two different screen will be used to isolate genes regulating Gpa3 or genes involved in the second pathway controlled by Gpa3: a two-hybrid screen with Gpa3 as a target and a genetic screen based on the phenotype of an overactivated Gpa3 allele. The genes identified in these screens will then be submitted to an extensive genetic and physiological analysis with the help of knock-out mutants to understand the regulation of the two pathways and the biological relevance of their coordination.
Training content (objective. benefit and expected impact)
This project will allow me to extend the knowledge and expertise I gained during my Ph.D. on signalling pathways and fungi to aspects of pathogenic development in fungi. Due to the growing interest of companies and of the academic field in fungal pathogens this will broaden my carrier oportunities for the future.
