Identification of genes involved in mouse cns development using gene trapapproaches and analysis of phenotypes generated by insertional mutations

Research objectives and content
I intend to use the gene trap strategy to identify novel genes involved in mammalian CNS development. The approach utilizes murine embryonic stem cells (ES) and a-gal-carrying reporter vectors containing a
splice-acceptor site. Upon integration of these vectors into introns a fusion transcript between the endogenous gene and the ,B-gal gene is generated. A gene recently isolated in the host laboratory using the gene trap methodology is involved in the general apoptosis pathway. The aim of this project includes the phenotypic analysis of the generated insertional mutation, and the functional characterization of the trapped apoptotic protein.
Training content (objective, benefit and expected impact)
I acquired a strong background in molecular biology, during several years of work on RNA processing in Xenopus laevis. I recently acquired technical and theoretical skills in mouse embryology. My objective is to broaden my knowledge in the transgenic mouse technology, and to use the gene trap approach to better understand CNS development and the role played onby apoptosis. A training of 24 months in the field would give me enormous career benefits and the possibility to carry out afterwards, in my country of origin, the learned techniques and know-how so far related to only very few labs.

Besides MPI, AMGEN Research Inc. supports the general direction of gene trap technology in the host laboratory. My project is linked to the establishment of the European Mouse Mutant Archive (EMMA) in Monterotondo (IT). Moreover, several clinical applications are linked to the characterization of the apoptosis factor I am working with (i.e.: cancer therapy and processes of inflammation)
