Functional characterisation of rhomboid

Objective



Research objectives and content The Drosophila rhomboid gene (rho) is involved in the activation of the EGF Receptor signalling pathway, leading to cell fate determination and differentiation at many developmental stages. Despite intensive genetic studies, the structure, function and partners of Rhomboid remain elusive. The aim of this project is to further our knowledge of the mechanisms by which Rhomboid leads to the activation of the EGF receptor. There are indications that Rhomboid may define a new family of proteins conserved through evolution, giving this project a broader impact. The novelty of my approach consists in its multidisciplinary nature, using biochemistry and cell biology to study the structure and intracellular localisation of Rhomboid, and molecular biology and genetics to perform two convergent screens that should enable the identification of genes and gene products that interact with Rhomboid and will help elucidate its function. training content (objective, benefit and expected impact) My scientific career will benefit greatly from the multidisciplinary approach to a problem that lies at the intersection of signal transduction,cell fate determination, and control of cell proliferation. This project also involves the use of multiple techniques that will be extremely useful in my future career. In particular, I will develop skills in Drosophila cell culture, and the use of the yeast two-hybrid system. I expect that the training gained during this project will greatly benefit my incorporation to the research system in Spain and also have a positive impact in other-researchers through future collaborations. Links with industry / industrial relevance (22) This project will have no direct industrial relevance,although it will further our understanding of oncogenesis and might therefore, contribute to the development of novel therapies.
