A structural investigation of the pigment binding sites in cp29, a minor antenna protein in higher plants

Objective



Research objectives and content Mutants of the plant light-harvesting protein CP29 will be generated, in order to identify and alter those residues hydrogen-bonding the prosthetic groups (chlorophylls). will be principal objective is to define the spatial orientation of each chlorophyll molecule, based on the atomic model of the related light-harvesting protein LHCIIb, and to determine the effect of hydrogen-bonding on spectroscopic properties of chlorophylls. Furthermore the mutants generated, along with a further set of mutants in which the strength of hydrogen-bonding interaction has been modified for one or more of the bound chlorophylls, will be used (in collaboration with a second European research group, led by Prof van Grondelle) to study excitation and energy transfer parameters. As well as the direct, structural information this will give on CP29 and related plant light-harvesting proteins, this will prove an invaluable model system in which to determine the factors effecting energy transfer in chlorophyll-proteins generally. Training content (objective, benefit and expected impact) - I will be trained in molecular biological techniques for the generation of recombinant and mutated polypeptides in E. coli, and in reconstitution of LHC polypeptides. I will also extend my current (limited) biophysical skills to in clude such specialised areas as pump-probe spectroscopy and fluorescence line narrowing.
