Phenotypic characterisation of nogo deficient mice.

Objective

Nogo has been shown to be a major inhibitor of regeneration in the adult CNS acting with a potency similar to MAG. Studies with the function blocking IN- 1 monoclonal antibody have allowed a limited investigation of the role of Nogo-A. Technical difficulties with antibody delivery, stability and specificity limited the scope of these studies. A definitive understanding of its full role can only come from the performance of lesions in animals in which the Nogo gene has been inactivated. We are generating such animals and will be in a unique position to study them. The successful candidate will utilize skills in the performance of spinal cord and other CNS lesions and monitor the growth responses of surviving neurons. Studies will also be necessary to quantitate and characterize the expression of residual Nogo-C expression and analyze possible compensatory changes in the expression of other inhibitory components of myelin. We have extensive experience in a wide-range of technologies and training will be provided in molecular-biological characterization of knockout mice, analysis of neurite-outgrowth, purification of inhibitory components from the CNS and expression of recombinant inhibitory proteins. Work will be carried out in the Neurology Centre for Excellence in Drug Discovery. This department has an excellent track record in the training and development of post-doctoral fellows that has access to some of the best research facilities, equipment, technologies and highly trained and motivated staff available anywhere in the world.
