Transgenic models for cardiovascular diseases

Objective

The Max-Delbrueck-Center for Molecular Medicine (MDC) in Berlin-Buch invites applications for EU-funded pre-doctoral fellowships (3-12 months) in the research area of functional genomics in the cardiovascular field. Research training will comprise the molecular biological analysis of genes and their regulation by the screening of libraries, the cloning of genes, the characterization of their structure, the evaluation of promoter activities, and methods for the isolation of DNA, RNA and protein from tissues, and gene expression measurements (Micro arrays, Northern blot, RNase-protection assay, RT-PCR, TaqMAN PCR, Western blot). The major focus of the training will be the generation of transgenic animals, i.e. techniques of embryonic stem (ES) cell culture and homologous recombination, microinjection of DNA in mouse and ratoocytes or of ES cells in blastocysts, and the cardiovascular phenotyping of animal models, i.e. the measurement of blood pressure and heart rate (tail-cuff, intra-arterial catheter, telemetry) for rat and mouse, of functional parameters of kidney and heart, of vascular contractility, of platelet aggregation kinetics, of basic biochemical parameters and hormone levels in plasma, urine and tissues, the icv injection of substances to study central cardiovascular control, and experimental models for cardiovascular diseases, i.e , aortic or coronary ligation to induce cardiac hypertrophy or infarction, respectively, induction of hypertension by angiotensin infusion, the two-kidney-one-clip-,or the DOCA-salt methods. Applicants must be registered in a PhD program and have the nationality of a Member or Associated State of the European Union except Germany. Women are encouraged to apply.
