Sperm-egg cell interaction and fertilisation in rice

Objective

The main objectives are as follows:
* To enlarge the knowledge of reproductive cells isolated from rice, with particular emphasis on their cellular and molecular aspects.
* To develop methodologies for introducing foreign DNA in sperm cells isolated from rice.
* To develop and standardize procedures for "in vitro" fertilization using isolated gametes.
* Construction of "gamete banks" for different varieties of rice plants.
* Regeneration of rice plants by "in vitro" fertilization using isolated gametes.
* Generation of transformed rice plants, with improved characteristics of agronomical interest.
Expected Outcome

The work carried out in this project should provide new techniques for gamete isolation from different rice varieties. The creation of a cDNA library of rice varieties and the identification of specific gene products for the sperm or egg cells is also expected. As a final goal the programme should lead to the introduction of genes of agronomic interest into sperm cells in order to obtain improved rice plants.
The key activities envisaged are:
* Detailed cytological and ultrastructural investigation on anthers, pollen grains and pollen tubes, stigma, style and embryo sac either by Transmission Electron Microscope or Confocal Laser Scanning Microscope.
* Establishment of experimental conditions for the isolation and culture of gametes.
* Isolation of mRNA from sperm cells and construction of cDNA libraries.
* Investigation on isolated male and female gametes with particular emphasis on their cellular, biochemical and molecular aspects.
* Development of "in vitro" fertilization techniques using isolated gametes.
* Investigation on fusion products (zygotes), with reference to the cytoplasmic organization.
* Transformation of gametes by insertion of foreign DNA.
* "In vitro" fertilization using transformed gametes.
