Coordination of plant oxidative enzymes as a key factor in degradation of organic xenobiotics

Objective

The initial stage of organic toxicants degradation in plants is their oxidative transformation. As it is well known, this is rate limiting in multi-step complicated process of detoxification. According to different considerations xenobiotic oxidation in plants is carried out via monooxygenase, or via peroxidase, or via phenoloxidase. On the basis of our investigations, we can suppose that none of the stated mechanism is not universal way for xenobiotic detoxification. Consequently, the hypothesis is drawn out, according to which, the interchange of oxidative systems in xenobiotic detoxification takes place, which should be determined by many internal and external factors.

The importance of this research is to determine exactly what enzymatic systems participate in the first step of xenobiotics oxidation - hydroxylation and to establish the mechanism of their coordination.

The goal of the research is the establishment of possible pathways of nitrobenzene oxidation in soybean and maize plants; the reveal of the fact of different oxidative enzyme systems interchange in xenobiotic hydroxylation.

To achieve the Goal following tasks will be addressed:

Effect of xenobiotic on cell ultrastructure.
Xenobiotic oxidation by monooxygenase, peroxidase and phenoloxidase enzymatic systems in subcellular fractions.
Quantitative input of each enzymatic mechanisms in xenobiotic oxidation.
The intensity of oxidation processes in relation with plant cultivation regime and age. Variants: etiolated plant seedlings; seedlings grown in light; seedlings infected with pathogenic fungus; seedlings grown on xenobiotic containing solutions; different age seedlings.

The expected results will contribute for more clear understanding of the mechanism of interchange, whether it is realized: by different oxidation enzymes interchange each other; by the modification of cytrochrome P450 activity; or by the combination of the two mechanisms.
