Objective
The overall objective of the proposed research is to collect data on the prevalence of porcine reproductive and respiratory syndrome virus (PRRSV) infection in the NIS, to isolate and characterize new PRRSV strains typical of these regions and to develop proper tools for differential diagnosis of infection. In order to achieve this objective, there is a need to establish reliable methods for virus isolation, to apply or develop new methods for the detection of PRRSV antigen and viral RNA in clinical samples and to develop serological tests for the detection of antibodies to PRRSV.
The technical approaches that are proposed are similar to those used in Europe, and this will ensure that the results will be compatible with most of the relevant data obtained in Europe and USA. In order to achieve the main objective, the following major steps are proposed:
To obtain preliminary data on incidence and distribution of antibodies against different genotypes of PRRSV in the participating NIS.
To characterize PRRSV strains currently circulating in the NIS countries at the molecular level and to study their epidemiological relationship to known strains from Europe, USA and Canada.
To use the above data to select a representative virus strain(s) and produce recombinant antigens of PRRSV isolates in a bacterial / baculovirus expression systems. These recombinant antigens will then be incorporated into a serological ELISA, the data from which will be used to determine the final configuration and validation of a novel serological tests against different genotypes of PRRSV among pigs in the NIS.
Despite the significance of PRRSV for the swine industry all over the world, very little is known about the spread of PRRSV within the NIS, and there is no information about PRRSV strains circulating in this area of the world. Data on the distribution of PRRSV in the NIS will help to complete the overall picture of PRRSV spread. The results will provide information to develop strain-specific discriminatory tests. As well as its significance for the NIS agriculture industry, the EU will also benefit from the results of this project, since new strains within the NIS could pose a significant threat to adjoining countries. The combined results of the project will also contribute to the control of this economically important disease, through the provision of accurate and sensitive serodiagnosis.
