Bacteriophage - a new approach for combating nosocomial respiratory infections caused by Pseudomonas aeruginosa

Objective

To minimize the risk of nosocomial respiratory infection caused by P. aeruginosa it is planned:
-to screen existing P. aeruginosa phage collection of the IBMV(GE) comprised of 30 clones against about 200 strains obtained from the IECS (GE) clinic;
-to isolate up to 20 new phage clones from environment;
-to select up to 10 candidate phases on the base of the their biological features and genetic character of the host-phage interaction;
-to investigate gene structure of the best 5 candidate phages with the purpose to avoid possible spread of imipenem drug-resistance;
-to perform clinical experiments comprising phage treatment of hospital environment of the Department of Lung Surgery in the IECS (GE);
-to elaborate a new inexpensive and efficient strategy based on application of specific bacteriophage(s) for prevention of P. aeruginosa nosocomial infection, which will result in a reduction in the cost of prophylaxis.

Research activities:

For realization of the goals of the project the following research activities shall be accomplished:

1) Microbiology studies of the clinical environment and patients;

2) Construction of the strain collection by using standard (host) strains from the ATCC and other collections and newly isolated and studied of nosocomial Pseudomonas strains;

3) Screening the strains for phage-sensitivity against the IBMV phage collection and phage clones isolated from environment during the present study;

4) Selection of candidate phage(s) and appropriate host(s);

5) Characterization of the candidate phages which involves the following studies:

a)Morphological and biological studies;

b)Study of the efficiency of plating (EOP);

c)Resistance of phages towards physical and chemical agents;

d)Serological relatedness of the phages;

e)Intracellular phage growth cycle (IPGC);

f)Ability of phages to transduce drug-resistance;

g)Characterization of the phage DNA and determination of the
genetic relatedness of phages;

6) Genetic studies of the host-strains which involves the following stages:

a) Detection of plasmids;

b) Detection of temperate phages;

c) Study of the frequency of occurrence of mutations to phage-resistance (PRM);

7.Extension of the host-range of the selected phages and creation of the complex phage preparations with broad spectrum of activity;

8. Experiments for assessment the prophylactic efficiency of Pseudomonas phage;

9. Analysis of obtained results and their statistical assessment.
