Role of phospholipases in the mechanism of cytokinin action

Objetivo

The objective of the project is to identify the role of different classes of plant phospholipases, namely PLA2, PLC and PLD, in the molecular mechanism of cytokinin perception and signal transduction. The background of the project is the recent finding by one of the research teams that the activity of PLD and to some extent of PLC are important for the in vivo realization of the primary cytokinin effect on gene transcription in different model plants. The work is planned to be undertaken on convenient and quantitative plant model systems (Amaranthus and Arabidopsis) established previously. This project seems to be unique not only because it brings first connection between phospholipases and cytokinin action, but also because its integrated character when all known phospholipases should be under study. The complexity of studied processes demands an interdisciplinary approach. Therefore our research platform includes experts in the field of plant molecular genetics, biochemistry (particularly phospholipase biochemistry) and plant physiology.

Teams from Czech Republic (I. Machackova, O. Valentova, I. Martinec) and Ukraine (V. Kravets) will study the early cytokinin effect on the activity of different classes of phospholipases: PLD, PLC and PLA2. The team from Moscow (G. Romanov) will accomplish pharmacological and kinetics studies of phospholipase activity in vivo. Many isoforms of phospholipases are Ca2+-dependent; therefore the team from S. Petersbourg (S. Medvedev) will perform a parallel analysis of calcium status during the cytokinin signal transduction. Also phosphoinositides PIP2 and IP3 which are widely involved in cross talk between different phospholipases ensuring their concerted action in signal transduction will be quantified (V. Kravets). Molecular analysis of gene expression and production of transgenic plants with altered phospholipase activity will be made by the team from Germany (T. Schmulling). Thus we have assembled research teams by which very complementary work can be carried out. At the end of the project we will identify phospholipase or different phospholipases whose activity is quickly affected by cytokinins.

We will determine the role of products of activity of this phospholipase(s) in the cytokinin-signalling cascade together with the role of calcium and phosphoinositides in this process. The presumable cross talk and concerted action of phospholipases belonging to different classes will be studied. Transgenic plants will be produced showing the biological role of defined phospholipases in cytokinin action and in overall plant growth. The expected results should help to decipher the pathway(s) of cytokinin signal transduction from recently discovered membranous cytokinin receptor CRE1 to promoters of primary responsive genes. This project could bring also new knowledge about possible functions and interaction of different plant phospholipases in the regulation of plant metabolism.
