Novel higher carboranes and boranes: synthesis and applications for antitumour neutron capture therapy, and new materials

Objetivo



The goal of this project is the synthesis of novel closo- and nido-polyboranes, carboranes and heteroboranes containing functional substituents bonded at carbon or boron atoms. The substituents may provide selective accumulation in tumours, or provide functional groups to allow incorporation into polymers with polyborane backbones. The substitutents will be systematically introduced, and the effect of isomeric substitutions elucidated. The main borane clusters which are the subject of the investigation in the project will be the higher closo- and nido-carboranes and boranes, including closo-1,2-C2B10H12, [B10H10]{2-} and [CB11H12]{2-} cages; nido-[7,8-C2B9H11]{-} [CB10H13]{-} and [B11H14] anions; and the aracho-anion [B9H14]. The substituents include: biological species such as sugars, aminoacids, nucleoside bases, ether lipids, and DNA intercalators; and simple functional substituents for polymer construction.

These compounds will be analysed for acute toxicity on small animals. Biological distribution of the compounds which do not have a high toxicity will be examined using in vivo mouse and rat tumour models.

Boron concentration in biological samples will be measured by the methods of prompt gamma-ray spectroscopy.
