Biophysical, biotechnological and chemical approaches to the investigation of the catalytic mechanisms of catalases, dioxygenase, laccases

Objetivo



This research project deals with the study of the structural, enzymological and regulatory properties of the electron-transfer paramagnetic metalloproteins - catalases, catechol and dihydroxybiphenyl dioxygenases, and lacases from native and mutant strains. The chemical derivatives of the above-mentioned proteins as well as their models will also be investigated.

The research activities will cover the following: isolation and purification of the above enzymes; preparation of their mutants (site directed mutagenesis), derivatives; their biophysical characterization (EPR, NMRD, UV- and CD- spectroscopies, kinetics, electrochemistry); 1H NMR studies of the proteins, their derivatives, adduct with substrates, ligands (conventional 1D and 2D SMR experiments); NMR studies of enzyme substrates, pseudosubstrates, inhibitors in the presence of the enzymes; and synthesis and characterization of the compounds which are structural, electronic and functional models of the enzyme active sites; and studies of the behaviour of these compounds in the reactions, which mimic the catalytic processes and which occur in the presence of the enzymes described.

As a result of this project it is expected that new valuable information will be obtained about the catalytic mechanisms of the proteins discussed and that it will be possible to postulate some of their structure-function relationships.
