Objective
This project concerns fundamental studies in bioenergetics and the regulation of biological processes and the clinical application of the results of these fundamental studies for understanding the mechanisms of pathogenesis of diseases and development of new methods of diagnosis. The fundamental studies of this project include experimental investigations of cellular respiration (oxydative phosphorylation) and cell function, for example the study of the contractile strengths by using the newly developed permeabilised cell and skinned fibres technique.
Also clinical studies of patients with cardiac diseases (myopathies, etc.) will be performed on the basis of modern NMR tomography and spectrometry available. By using these methods the energy and metabolic fluxes in different cells of different organs in the body will be studied and quantitatively analysed on the basis of the results of the experimental and theoretical research. As a result of this project it will be possible to identify new pathogenic mechanisms. These results will be beneficial for the development of new methods of diagnosis.
The project will also analyse several classes of pharmaceutical agents by using several novel techniques.
This project builds on an intensive international co-operation between the participating laboratories over the past years. This co-operation has already produced numerous publications. Recently several of these laboratories have been included into the reseaux formation research programme which includes joint research projects on the problems of control of oxidative phosphorylation, its modelling, and analysis from the biothermokinetics point of view. Thus the theoretical part of this project will build on the solid foundation of a well-established European research network, which will guarantee its high efficiency and level.
