Objective
The general objective of this project is to study new aspects of the molecular regulation of cholesterol metabolism by orphan nuclear receptors and the contribution of the underlying mechanisms to cholesterol homeostasis. To achieve the objective of this application we will pursue the following specific aims: 1. Analysis of the role of orphan nuclear receptors on the regulation of the expression of genes involved in cholesterol metabolism. 2. Systematic identification of novel target genes regulated by orphan nuclear receptors. 3. Determination of the crystal structure of the legend-binding domain of HNF-4, CPF, SHP and COUP-TFII. 4. Search for legends for HNF-4, CPF, SHP and COUP-TFII. 5. In vitro and in vivo analysis of the newly identified legends. This project will allow finding new drug targets for novel treatments of hypercholesterolemia to lower the risk of developing atherosclerosis an the incidence of cardiovascular diseases associated with this metabolic disorder.
