Mitochondrial biogenesis as a cause for leigh syndrome

Objective

Innovative approaches will be used to achieve the main objectives:
a) To identify new factors involved in COX assembly, Tandem Affinity Purification with mass spectrometry and micro arrays technology will be used
b) To study in vivo assembly and turnover of cytochrome c oxidise and determine the role of SURF-1, two- dimensional electrophoresis with metabolic labelling and a novel approach based on the use of fluorescent tags (FRET) will be applied
c) micro arrays will also be used to compare expression levels of the genes in the genome of wild-type, DeltaShy and DeltaShy revertants in different physiological conditions and d) approaches developed in yeast will be tested for diagnostic and information obtained from spontaneous revertants will provide important indications for therapy. These innovative strategies in combination with the high-quality research of the host group will add strong technical background to the applicant and will ensure a successful postdoctoral stay.
Considering the background of the candidate in the mitochondrial field and her motivation, her stay in the lab will be also beneficial and stimulating for the research of the resident lab community.
