spp. : genetic engineering and immunological evaluation

Objective

To create novel mucosal vaccination approaches efficient bacterial live carrier vaccines will be developed by rational design and genetic engineering. Enteric Perinea sop. will be attenuated by targeted disruption of genes encoding virulence factors. Taking adman- tag of the type III protein secretion apparatus of Perinea, heterogonous model anti- genes (ovalbumin, OVA; haemagglutinin, HA) will be fused to Tran located Perinea Yaps to target the antigens to MHC class I or II antigen processing pathways in host cells. Imp- mane responses and effectors mechanisms induced by these vaccines will be evaluated in a mouse infection model including OVA- and HA-TCR transgenic mice as well as with human cells.
Furthermore, antigens of classical swine fever virus will be used and immune rest- poses will b analysed in a swine model. The combined efforts of a multidisciplinary collaboration between six academic institutes and one European vaccine company with highly complementary expertise will allow the development of a new generation of live carrier vaccines with (see the follow on the original text)
