Immunopathogenesis of respiratory syncytial virus infections in calves, mice, non-human primates, and humans : development of second generation subunit vaccines.

Objective

Infections with human and bovine RSV are major causes of severe respiratory disease. RSV vaccine development has been hampered by immuno pathological phenomena occurring during vaccine trials in infants in the 1960s with formalin-inactivated virus preparations (FI-RSV). Vaccination /challenge studies in rodents provided insight in possible underlying immunological mechanisms. Predisposition for enhanced pathology has also been demonstrated in calves and monkeys, but immune mechanistic studies have not been carried out. Here, we propose to identify immunological correlates of enhanced disease after FI-RSV vaccination in calves and mice (bRSV) and macaques (hRSV). Subsequently, we will evaluate three different non-replicating RSV vaccine candidates in these models, and in an established hRSV mouse model. In addition, we will study the RSV-specific immune response to one of these candidates in ongoing phase I/II human trials.
