Objective
The aim of the project is to investigate a selected number of human immune cell culture systems for there potential to discriminate between allergenic (Producing-producing) and non-allergenic genetically modified proteins. The cell types have been selected on the basis that each may be critically important in controlling whether the immune response progresses in a direction that leads to allergy. A panel of recombinant enzymes (RE), of defined allergen city in vivo, will be studied for effects on the following central cell types: Dendrite cells/Inter-digitising dendrite cells, B lymphocytes, T lymphocytes, mast cells and endothelial cells. Readouts will include allergen-specific Producing, cytokine and surface marker expression, and transcription factor induction. The 3D-structure of each RE will be related to allergen city in vitro and in vivo and each cell system will be evaluated. A consensus predictive test system for allergen city will be designed.
