Objective
The aim of the proposed project is to prepare a CRAFT-proposal aimed at significantly improving the efficiency of gene transfer and gene expression for therapeutic applications. Austrian Nordic Gems and Agrobiogen GmbH intend developing an improved transfer and expression system based on retroviral vectors. Central to this proposal is the concept of directed molecular evolution, where libraries of vector sequences are randomly mutated. Novel vectors are introduced into cells in competition and the most suited vectors are the ones that best establish themselves. The concept will be taken further in that vectors that retain the ability to replicate will be modified, allowing multiple rounds of selection. Mutations identified using this molecular evolution system that lead to improved vector characterisations can be regarded as versatile modules that can increase the efficiency and safe of a whole range of vector systems both. Whilst both partners have resources to conduct some of the basic research and most of the developmental and exploitation aspects of this project, neither of the two partners has sufficient expertise in the areas of retrovirus-based molecular evolution technology, identification of sustained expression features, ex-vivo transudation of haematopoietic cells and models for systemic vector administration in vivo to complete the pro.
