Dynamics and composition of filamentous micro-organism communities in industrial water systems

Objective

Filamentous microorganisms cause bulking sludge resulting in major problems in industrial wastewater treatment plants (Wets). Development of prevention strategies is hampered by lack of identification methods for these organisms. Thefilamentous biodiversity in a large number (>100) industrial water systems is explored using conventional and innovative molecular identification techniques. Filaments are micro manipulated from activated sludge and species pacific 16S rRNAsequences are derived via PCR or reverse transcripts PCR.Probes and primers for respectively Fluorescent In Site Hybridisation and PCR and multiplex quantitative are designed to monitor filament population composition and dynamics. Filamentecophysiology is studied both in pure culture and in site using microautoradio-graphy. This study will result in a methodology to study biodiversity and dynamics of filamentous microbial populations in industrial Wets.
