Objective
The complete sequencing of microbial organisms is producing a revolution in both basic science and in the biopharmaceutical industry. A broad range of novel targets for therapeutics, and new biological functions are being discovered. DNA sequencing, nor the high throughput analysis of gene products did not design current gene manipulation technologies for the broad range of microorganisms that are being characterised. The enormous amount of new information requires a new generation of genetic tools for the analysis of genes and their mutants, and the characterization of their products by heterogonous over expression of open reading frames (Orbs). Using stably inserted and tightly regulated circuits, we aim to construct a collection of new, specialized strains and genetic tools, which can be used to engineer conditional phenotypes and to facilitate genetic manipulation. New means to improve recombinant protein expression and genomic engineering in a broad range of bacteria will be developed.
