Objective
This project addresses the question of interactions between PAHs in the urban ambient air mixture and how they may affect the hypothesis of risk additivity which underlies current risk assessment methodologies for PAH mixtures. Studies in vitro (using rodent and human tissues) and in vivo (rodents) are proposed to examine qualitatively and quantitatively parameters related to the tumour initiating and promoting potency of selected PAHs, acting alone or in mixtures, and to test the hypothesis of risk additivity. The parameters to be examined include, among others, macromolecular binding, in vitro and in vivo mutagenesis, changes in gene expression, receptor binding, and others, as well as initiating and promoting potencies in the 2-stage mouse skin test. The outcome of the project may aid in the definition of improved air quality standards and monitoring strategies.
