Objective
The design and construction of binuclear copper centre based biosensors and biocatalysts are the two objectives of this project. Toward these aims we propose to investigate the molecular basis of reversible oxygen binding and activation as observed in the active site of structurally homologous binuclear copper proteins that physiologically function either as oxygen carrier or as selective monooxygenases and catechu oxidises. The objectives will be achieved by inclusion of these proteins into optically and chemically inert solid matrices. This will allow our industrial partners to construct a low cost optical oxygen biosensor for biomedical use and will provide a selective, removable biocatalyst for the conversion of endogenous phenol substrates in food processing. The available genetically engineered systems will allow for the tailoring of sensitivity and specificity of the proteins used to the implementation demands.
