Objective
To address the role of mRNA structure and stability in controlling gene expression in yeast a novel modular mRNA, based on the
Saccharomyces cerevisiae PYK1 (pyruvate kinase) gene, will be synthesised to facilitate in vitro manipulation of both coding and non-coding sequences. Three particular aspects of the problem will be addressed : (1) the role of mRNA primary and secondary structure in determining translational efficiency paying particular attention to the 5' untranslated region (5'UTR) and the importance of codon bias; (2) the relationship between the primary and secondary structure of an mRNA and its stability; and (3) the development and exploitation of an in vitro mRNA degradation assay.
This proposal deals with an Italian-Dutch twinning project with the aim to find support for an effective scientific and technical interchange in the field of laser induced "on-line" diagnostics used in the study of laser assisted photochemistry. The Italian
photochemical groups will benefit from the experience of the Dutch group in developing advanced spectroscopic tools; the Dutch, on the other hand, are eager to develop new intrumentation for dedicated purposes. Cooperation will speed up industrial application of laser both in material processing and in unique space- and time-resolved diagnostics. Mutual interchange of information, obtained with complementary techniques, will also allow to rapidly understand mechanism of each process and to drive it optimizing all the parameters.
