Objective
Peptides corresponding to defined segments of rat thyroglobulin will be synthesized in E. coli. The in vitro synthesized polypeptides will be submitted to iodination under conditions favoring iodination and/or iodination plus coupling. The iodoamino acids distribution of the different fragments will be determined, the peptides containing thyroid hormones will be puridied by HPLC and sequenced to establish if hormone synthesis occurs at the same tyrosines used in vivo. Both structural and mutagenic analysis of such fragments should allow to deduce the minimal requirements for thyroid hormone biosynthesis.
