Objective
Two new approaches will be developed for the characterization of primary photosynthetic reactions by vibrational spectroscopy. Firstly, using recent advances in protein spectroelectrochemistry, the changes of redox states of the cofactors within the bacterial RC will be driven electrochemically and the bonding interactions of these cofactors will be assessed by FTIR spectroscopy. Secondly, time-resolved IR and FTIR spectroscopy for transient species decaying in the ms to micro-s range will be applied to RC of both plants and bacteria. Kinetic IR spectroscopy will provide access to the molecular details of the intermediates involved in the primary reactions. These two approaches will contribute to characterize the dynamics of primary electron transfer and the structural role of cofactors-protein interactions in the efficient stabilization of solar energy.
