SEQUENCE ANALYSIS AND CLONING OF NON CATALYTIC SUBUNITS FROM ACETYLCHOLINESTERASE ASYMETRIC FORMS

Objective



The asymmetric forms of vertebrate cholinesterases present an original structure, in which the catalytic subunits are assembled with non-catalytic peptides, constituting notably a triple helical collagenic tail, and their expression reflects a specialized differentiation of muscular and nervous cells. We plan to analyze the structure of the non-catalytic subunits of Torpedo and Electrophorus asymmetric acetylcholinesterase by microsequencing, to synthesize oligonucleotide probes for cloning their cDNAs, and prepare polyclonal and monoclonal antibodies. In a second period, we wish to clone the tail subunits of avian and murine cholinesterases, and thus obtain tools to study their biosynthesis.
