Towards complete physical map of the drosophilia melanogaster genome - the second chromosome

Objective



This proposal is essentially a continuation of the Genome mapping programme funded by the EC under Contract * STP2-0477-C(TT). Here we propose to continue our mapping effort, that started with the X chromosome, into the
autosomes. While the basic strategy is the same, some modifications have been incorporated aiming at obtaining faster and more reliable results. In addition to obtaining cosmid contigs we will incorporate in the present study the use of YACs and P1 clones to link contigs, taking advantage of the
larger size of the inserts of these vectors. Also, a crucial point of the proposed study is the definition of sequence tagged sites through the sequence determination of the ends of canonical cosmids. These STSs will allow a faster dissemination of possible entry points for cloning of genes
by laboratories of the Drosophila community. The next items addressed by this proposal deal with the localization of known genes through hybridization studies, the mapping of the heterochromatic regions of the chromosomes (approximately 20% of the genome) by collaborating with various groups that that are active in this field, and by improving software related problems
(including the issue of databases). Finally we will address the problem of the distribution of DNAs among the Drosophila community, a problem that will ultimately be resolved when enough STSs have been obtained.
