Objective
Chemical disinfectants and antiseptics are used as aids in the prevention of cross-infection in a wide variety of areas and for a broad range of applications.
In the project of European Directive on Biocidal Products, efficacy data of disinfectants are required for placing these products on the market. The annex Vl of this project of Directive specifies that these efficacy data can be established by using European Standards.
However the present standards (EN 1275, EN 1276 and Pr EN 1657) dealing with test methods for the evaluation of anti-microbial activity of disinfectants and antiseptics prepared by CEN/TC 216 << Chemical disinfectants and antiseptics )) are the result of compromises between the different experiences gained by some European countries through their national standardization and require some research and validation in order to make sure that they can really serve as reference documents for application of the Directive.
Indeed, for example:
- EN 1275, EN 1276 and Pr EN 1657 propose two alternative methods: thedilution-neutralization method and the membrane filtration method while noextensive comparison between the alternative methods, when used in theconditions stated by the standards, exist. In addition, an alternativemethod combining neutralization and membrane filtration has recently beenproposed and may be suitable in order to avoid some limits of the membranefiltration method as described in the current standards,
- In Pr EN 1657, two alternative counting techniques are proposed (the pourplate technique and the spread plate technique) and again no comparisonexists between them in the condition stated in the standard,
- In the three standards a range for the initial count of extensive strainsis given but its method of determination is left open. This constitutes acritical point since it has been proven difficult in certain cases to obtainan initial count fitting in the determined range because of the countingtechnique.
Therefore this project intends to validate the test methods and to confirm that some alternatives proposed in the test methods are interchangeable and yield the same efficacy results. This project also intends to give evidence of the interest of the alternative method combining neutralization and membrane filtration.
For this purpose five work packages are planned with first a preliminary test to determine the range of suitable dilutions of the chosen products, then the testing on the basis of the three standards by 20 laboratories and a statistical analysis of the results.
