Objective
We are proposing to compile a complete physical (molecular) map of the genome of Drosophila melanogaster. This will entail the "fingerprinting" of ca. 30000 DNA fragments (each 30-40 kbasepairs; cloned in a cosmid vector) and their assembly into an ordered array. The project will follow the methods developed in the pioneering work of Sulston and collaborators on Caenorhabditis elegans, and will also take advantage of a special feature of flies, the polytene chromosomes. The physical map wil be integrated with available genetic and cytogenetic information into a Database, which, together with the ordered cosmid library, will be of enormous value to all Drosophila workers.
