Protein quality in barley

Objective

The aim of the project is to identify ways to improve the nutritional value of barley seed protein without reducing the grain yield.
Though several cereals may be produced in excess within the European Community (EC) their protein content and quality, mainly determined by their amino acid composition, leads to a situation where still considerable import takes place. In this situation imporvement of protein quality is highly required. To some extent this can be attained by balanced fertilizer applications; potentially more progress may be expected from plant breeding programmes.

An attempt was made to replace part of the nutritionally inferior storage protein hordein by proteins with a better amino acid composition. Some crosses have been made, but results have not been indicated.
It is attempted to replace some of the nutritionally inferior storage protein, by storage proteins with a better amino acid composition. Different high-lysine storage proteins present in small amounts in the barley seed are identified, and genetic methods to duplicate the structural genes coding for these proteins are developed.
The project is an integrated part of a long term programme with the aim specified above.
It has been shown that the barley seed contain two lysine-rich proteins, Beta-amylase and protein Z, which apparently acts as storage proteins, Bmyl and Pazl, were located to chromosome 4 in studies employing barley chromosome addition lines in wheat. Further, we have shown that Pazl is located near the centromere of chromosome 4, while Bmyl is located distal on this chromosome. Therefore, Pazl is best suited to be duplicated by crossing selected translocation lines.
A large number of lines having translocations between chromosome 4 and other chromosomes are studied to find lines with translocation breakpoints near Pazl. Crossing of translocation lines with this gene one or more times. Finally, the effect of the duplication on the amount of protein Z and protein quality can be evaluated.
