Objective
14-3-3 proteins play a role in many cellular functions: they bind to an regulate several proteins which are critical for cell proliferation and differentiation. Most notably 14-3-3 interacts with and potentially regulates Raf-l, Bcr and protein kinase C. The crystal structure of 14-3-3 has recently been solved at NIMR and it is now important to determine the structure of this protein in a complex with protein kinases. The specific objective of this proposal is to determine the atomic structure and the mechanism of the interaction with Raf-l, a Ser/Thr kinase which plays a key role in the MAP kinase cascade. The project involves the expression and purification of various Raf-constructs and the biochemical and biophysical characterization of the 14-3-3/Raf interactions and the subsequent crystallization of stable 14-3-3/ Raf-l complexes.
