Objective
The success of vaccines designed to fight infectious diseases is largely dependent upon knowing the effectors mechanisms responsible for protection. Moreover, as a consequence of the new technologies employing poorly immunogenic synthetic and recombinant antigens, the identification and characterisation of efficacious adjuvant has become crucial for the development of successful vaccination strategies. The project foresees the following objectives:
1) To clarify the role of newly described regulatory patterns of cytokine production by macrophages in innate and adaptive immune responses;
2) To identify natural factors regulating the development and life span of DC, which may be relevant for their in vivo generation and survival;
3) To evaluate the impact of selected soluble mediators on the capacity of DC to bridge innate and adaptive immune responses;
4) To develop animal models suitable for testing the adjuvant capacity of cytokines and of DC-based immunisation protocols. The overall results of the studies described in this project are expected to yield new and important information, relevant for the development of novel and more effective immune interventions.
