TARGETING TUMOUR-VASCULAR/MATRIX INTERACTIONS

Objective

This project intends to explore and validate new targets and pathways for innovative treatment of solid tumors directed towards tumor vascular matrix interactions, define validated targets towards tumour neovessel formation and create effective and tumour-selective strategies that complement or subsitute established therapies. The apparent lack of success in translating angiogenesis research into the clinic is in our view based on the fact that the research field is fragmented, and no standardized tools and models have been identified that reliably reflect the complexity of tumour angiogenesis mechanisms in humans. This integrated project will identify and validate new therapeutic targets directed towards tumor vascular-matrix interactions, develop new therapeutic strategies and implement such strategies in pre- and clinical trials. The project will represent a virtual research institute ine Europe and consists of 14 higly competent research centers within the field of angiogenesis research. To define and validate new targets related to the tumour vascular transcriptome and proteome, the consortium will etablish high throughput functional screening technologies for the identification of novel secreted factors that regulate endothelial cell growth and survival. This includes the use of robotics platforms that will be used to identify cDNAs with specific cellular functions. In this application, basic science, translational research and clinical activities are strongly intergrated, in order to validate defined targets and new therapeutic principles for clinical implementation.
