New and Environmentally Friendly Methods for Making Compounds with Medicinal Importance: Novel Singlet Oxygen Chemistry Combined with Cascade Reactions for the Synthesis of Bioactive Natural Product

Objective

In accordance with the goals of the Marie Curie scheme set out in the commission¿s Work Programme documentation, the project, which is the subject of the current proposal, seeks to fulfil its primary objectives through the vehicle of a state-of-art laborat ory-based research project falling within the domain of synthetic organic chemistry. The primary objectives are the further development of the international career prospects of the individual applicant, Dr. T. Montagnon, augmentation of her research manage ment skills, and enhancement of her cutting-edge specialist skills. The two-year research project, outlined in detail herein, is a groundbreaking and highly promising approach towards the synthesis of a disparate series of bioactive natural products (pruno lides A-C, premnalane A, stolonic acids A and B, and stolonoxide A-D). It seeks to employ the internationally recognised excellence of the Department of Chemistry (University of Crete) in singlet oxygen chemistry, in combination with, the established exper ience of the scientist-in-charge, Prof. G. Vassilikogiannakis, in the field of natural product total synthesis, to expediently achieve its aims. The synthetic programme will implement novel, efficient, and leading-edge strategies to synthesise the target c ompounds. It is anticipated that powerful new methods will be developed from this work that will then have general applications in synthetic chemistry. Subsequent to the synthetic chemical investigations, the medicinal potential of the natural products, an d carefully designed analogues thereof, will be explored through the established international collaboration between Prof. G. Vassilikogiannakis and Prof. H. Fong (Department of Pharmaceutical Science, University of Chicago, U.S.A.). Three disease areas ar e to be targeted - cancer, HIV, and malaria.
