Objetivo The mammalian phosphoinositide (PI) 3-kinase-related kinase mTOR (mammalian target of rapamycin) occupies a central role in a signalling pathway involved in the control of cell growth and proliferation. It integrates signals arising from nutritional cues as well as hormones/growth factors to regulate growth by altering translation and transcription programs.Not surprisingly, alterations in this pathway has been directly linked to a number of disease states including cancer. The host laboratory has recently discovered a novel evolutionary conserved protein, termed URI, that operates downstream of TOR in both yeast and mammalian cells to contribute, directly or indirectly, to rapamycin-sensitive gene expression.In this proposal, we aim at- elucidating the function(s) of URI in cell growth and proliferation in mammalian cells,- determining the significance of rapamycin-sensitive phosphorylation of URI in modulating its function in growth control and gene expression and- identifying novel interaction partners of URI. The proposed studies will provide new leads towards a clearer understanding of URI function(s) in cell growth and proliferation and the mechanism underlying the control of cell growth and gene expression by mTOR.
