Tools for integrated studies of the immunoregulatory properties of commensal microbiota

Objective

The infantile microbiota of Western societies is deficient in microbes that support development of tolerance to innocuous antigens, thus predisposing to allergy development. Infantile populations from non-Westernized countries need to be examined to identify protective microbial exposures for the developing immune system.
The purpose of this TOK is to build a strong method- and knowledge-based platform focussed on identifying microbes with immunoregulatory properties in the infantile gut flora and testing their mode of action ultimately aiming at rational design of probiotics with desired immunoregulatory properties. For this purpose, we will transfer methodological knowledge between the applicant and two partners in the Associated States Romania and Turkey to permit development of a common protocol for studies of infantile gut microbiota, immune development and protection from clinical allergy. In addition, the non-culture dependent technique T-RFLP will be modified and developed for assessment of infantile microbiota composition in birth-cohort studies. This involves building a data-base from known T-RFLP patterns in combination with culture and sequence data for intestinal bacteria deriving from both Westernized and non-Westernized infantile populations. Further, tools for studies of the effect of selected microbes on the immune system will be advanced and methodology for systematic testing of the immunoregulatory potential of selected microbes in humans will be established in cooperation with a probiotic SME. The project aids in defragmenting the European Research Area and increase its competitiveness by integrating researchers from EU member and Associated States, basic and clinical scientists and academic and company-driven research.
