Mid-Term Report Summary - LIGHTDRIVENP450S (Light-driven Chemical Synthesis using Cytochrome P450s)

In this research initiative we aim to establish solar light driven production of high value diterpenoids using moss and cyanobacteria as production hosts. Focus is on the two diterpenoids forskolin and ingenol-3-angelate. The genes encoding the enzymes catalyzing the two pathways from geranylgeranyl diphosphate will be identified and functionally characterized by transient expression in tobacco or yeast. Using targeting sequences, the entire pathways for the diterpenoids will be transferred to the thylakoid membranes of the photosynthetic hosts. By introduction of optimal electron carrier proteins, solar light driven electron transfer from photosystem I will be channeled directly to the cytochrome P450s. The envisioned commercial production system to be developed for production of forskolin and ingenol-3-angelate can easily be adapted to production of a multitude of other high value diterpenoids. The production is driven by solar light with carbon dioxide from the atmosphere as the sole carbon source.
