THE GIANT PROTEIN TITIN: AN INTEGRATED APPROACH TO MOLECULAR CHARACTERISATION OF THE GENE AND ITS PRODUCT

Objetivo



Titin is the largest polypeptide yet described and an abundant constituent muscle. Its function and properties are to be investigated in an integrated study at both the protein and gene levels. At the protein level, X-ray diffraction of ordered fibres of purified titin will provide information about the folding of the polypeptide chain and the molecular mechanism of shape changes. At the nucleic acid level, the cloning will allow the analysis of the structure of the gene and its transcriptional organisation. The exact locations of particular sections of sequence will be determined in labelling studies using relation of titin to other proteins and its possible involvement in genetic diseases to be explored.
